Driven by the immunoglobulinenhancer elements, MYC is constitutively<\/p>\n<p>expressed. DNA demethylation of BLcell lines is known to repress B-cell<\/p>\n<p>phenotype, whereas the mechanisms and itsconsequences are to be elucidated. We<\/p>\n<p>found that decitabine(5-aza-2-deoxycytidine, 5-aza-dC) treatment led to<\/p>\n<p>down-regulation of MYCexpression and inhibition of proliferation, which was<\/p>\n<p>associated with therepression of immunoglobulin enhancer elements and aberrant<\/p>\n<p>expression of E2A and ID2. We showthat decitabine activates NF-kB, thereby<\/p>\n<p>upregulating ID2, a novel NF-kB targetgene, which partly expains the loss of B<\/p>\n<p>cell phenotype. We conclude that DNAmethylation patterning of BL cells sustains<\/p>\n<p>the constitutive expression of thetranslocated MYC oncogene bymaintainance of B<\/p>\n<p>cell phenotype; and interference with the epigenetic program by decitabine<\/p>\n<p>results notonly in alteration of the specific differentiation program but also<\/p>\n<p>compromisedoncogene expression.
